A protein molecule can undergo irreversible structural rearrangements in response to thermal treatment and their consequences can be investigated by fluorescence spectroscopy, inactivation kinetics and molecular modeling. Using this approach, the conformational dynamics of ALP was studied in the selected temperature range from 25 to 70 degC.
The results showed that the heat treatment induced slight changes in the overall structure of ALP with an increase in helix contents up to 30 and 33 % and decrease of beta turns motifs up to 45. Moreover, ALP was found to be rather stable against denaturing thermal stress due to the presence of two disulfide bridges (Cys121-Cys183 and Cys467-Cys474), which can stabilize the protein structure and protect it against thermal perturbations.
At the same time, fluorescence spectroscopy investigations demonstrated that ALP was able to efficiently quench its own intrinsic fluorescence with both acrylamide and KI. A spectral contour map of the three-dimensional projection of the ALP three-dimensional structure displayed two peaks with the location of peaks 1 and 2 being mainly determined by the intrinsic fluorescence of Trp and Tyr residues. This information was also confirmed by Stern–Volmer analysis.
